SMAD4(wild type)
SMAD4 is important for cartilage homeostasis. The inhibition of SMAD4 can induce chondrocyte apoptosis and aggravate osteoarthritis progression. Based on the online repository of miRNA targets, SMAD4 might be a target of miR-3074-5p in the course of osteoarthritis.
We constructed a portion of the gene encoding SMAD4. By inserting its 3’UTR region (414bp) behind the luciferase gene in the vector, we could determine the expression level of this gene segment. This fusion expression does nearly no harm to the normal physiological state of firely luciferase.

Design Notes
Based on the online repository of miRNA targets, SMAD4 might be a target of miR-3074-5p in osteoarthritis. Here we designed the base sequence of the base segment binding to 3074 as well as the segment of its upstream and downstream
Source
The base sequences were derived from the database of targetscan website.
